Sometimes when I click on SESSION è NEW CONNECTION I get the following
If I click OK the new session screen comes up but I can only use connections
that are in its history.
The TNS list seems empty.
Does this mean someone is editing it?
Yes. Some other application has it locked exclusively and is not allowing other
applications to even read it. Word is such an application that does this. In the
past an error message was not shown and users had the incorrect impression that
no error meant all is good. This has come up through support recently. Toad 10
exposed the issue with an exception dialog. Toad 10.5 has a friendlier message I
think.

Roughly the same message in the latest 10.5 beta:
Cannot open file
"C:\Oracle\product\10.1.0\Client_1\Network\Admin\tnsnames.ora". The process
cannot access the file because it is being used by another process
